<title>Aurora B Kinaz İnhibitörü BI 831266 Molekülünün Sisplatin Tedavisinin Sitotoksik Potansiyeli Üzerine Olan Etkisinin Belirlenmesi</title>

ÖZET:&#13;
Kanser, proteinlerin işlevsizliği nedeniyle hücrelerin kontrolsüz bir şekilde çoğalması sonucunda vücutta meydana gelen tedavi edilmesi zor bir hastalıktır. Kanser tedavisinde kullanılan konvansiyonel yöntemler başarılı sonuçlar sağlasa da bu tedaviler hastalar üzerinde ciddi yan etkilere ve bir süre sonra hücrelerin bu tedavilere direnç geliştirmesine neden olmaktadır. Bu önemli nedenlerden dolayı kişiselleştirilmiş tedaviler oldukça önem arz etmektedir. Kemoterapide hali hazırda kullanılan ilaçlarla küçük molekül inhibitörlerin kombinasyon tedavileri sinerjistik etki yaratarak bu olumsuz etkileri ortadan kaldırmak için önemli rol oynamakta ve küçük molekül inhibitörleri önemli kılmaktadır.&#13;
Bu çalışmada, hücre döngüsünün önemli bir regülatörü olan Aurora B kinaz inhibisyonunun konvansiyonel anti-kanser tedavisinde başarılı bir şekilde kullanılan sisplatinin sitotoksisitesi üzerine olan etkisi araştırıldı. Aurora B inhibisyonu için daha önce üzerine herhangi bir kombine sitotoksik analiz yapılmamış olan BI 831266 molekülü kullanıldı. Sisplatin ve BI 831266 molekülünün mono ve kombine tedavi sitotoksik etkileri insan meme kanseri hücrelerinde (MCF-7) incelendi. MCF-7 hücreleri artan dozlarda sisplatin ve BI 831266 molekülü ile tedavi edildi ve sitotoksik etki koloni sağ-kalım analiziyle belirlendi. IC50 dozları belirlenen sisplatin ve BI 831266 molekülünün sinerjistik etkisi kombine tedavi ile belirlendi. Sisplatin ve BI 831266 molekülünün kombinasyon tedavisi, koloni sağ-kalım analizi ile kanser hücrelerinin koloni oluşma kapasitelerini inhibe ettiği gösterildi. Sisplatin ve BI 831266 molekülünün mono ve kombinasyon tedavileri MCF-7 hücrelerinde DNA hasarı ve apoptotik sinyal yolaklarındaki proteinlerin ekspresyon düzeylerine western blot yöntemiyle bakıldı.&#13;
Bu çalışmada elde edilen veriler, sisplatin ve BI 831266 molekülünün kombinasyon tedavileri hücre canlılığını azalttığı ve koloni sağ-kalımlarını baskıladığı, apoptozisle ilişkili proteinlerin ekspresyon seviyelerini regüle ederek DNA hasarına ve apoptoza sebep olduğunu göstermektedir.; ABSTRACT:&#13;
Cancer is a difficult to treat disease that occurs in the body as a result of the uncontrolled proliferation of cells due to the dysfunction of proteins. Although conventional methods used in cancer treatment provide successful results, these treatments cause serious side effects on patients and after a while, the cells develop resistance to these treatments. For these important reasons, personalized treatments are very important. Combination treatments of drugs currently used in chemotherapy and small molecule inhibitors play an important role in eliminating these negative effects by creating a synergistic effect, making small molecule inhibitors important.&#13;
In this study, the effect of inhibition of Aurora B kinase, an important regulator of the cell cycle, on the cytotoxicity of cisplatin, which has been used successfully in conventional anti-cancer therapy, was investigated. For Aurora B inhibition, BI 831266 molecule, on which no combined cytotoxic analysis was performed before, was used. The mono- and combined-therapy cytotoxic effects of cisplatin and BI 831266 were investigated in human breast cancer cells (MCF-7). MCF-7 cells were treated with increasing doses of cisplatin and the BI 831266 molecule, and the cytotoxic effect was determined by colony survival analysis. The synergistic effect of cisplatin and BI 831266 molecule, whose IC50 doses were determined, was determined by combined treatment. Combination treatment of cisplatin and BI 831266 molecule was shown to inhibit colony formation capacity of cancer cells by colony survival assay. Mono and combination treatments of cisplatin and BI 831266 molecule DNA damage and expression levels of proteins in apoptotic signaling pathways in MCF-7 cells were evaluated by western blot method.&#13;
The data obtained in this study show that combination treatments of cisplatin and BI 831266 molecule reduce cell viability and suppress colony survival, and cause DNA damage and apoptosis by regulating the expression levels of apoptosis-related proteins.

<title>The Effect of Curcumin on P53 /TIGAR and Autophagy Pathway in Human Osteosarcoma U2OS Cell Line</title>

Osteosarcoma is a cancer disease from primitive bone tumor initiation and evolution of tubular long bones include femur, humerus and tibia. Osteosarcoma disease is defined and diagnosed throughout the inspection of tumor osteoblast cells that produce osteoid. Turmeric (Curcuma longa) is the most effective component in dried ground rhizomes of a member Zingiberaceae family. Curcumin have excellent free radical scavenging properties, however curcumin is almost the strongest antioxidant present in turmeric. Turmeric seasonal herbaceous plant with long lateral ramifications originally comes from Southeast Asia. Curcumin properties include anticancer, anti-bacterial, anti-inflammatory and antioxidant. Curcumin as chemical therapeutic agent demonstrated to suppress or inhibit various signaling pathways includes arrest tumor cell proliferation, inducing autophagy or apoptosis, inhibit malignant cell invasion and suppressing inflammatory responses. Despite the fact that effects of curcumin in cancer has been broadly reported, but the weather of curcumin on p53/TIGAR and autophagy has not been addressed in osteosarcoma U2OS cell line. In this present study, we screened the weather of curcumin after treated U2OS cells with curcumin, cultured cell were treated by various concentrations (10, 15, 20 and 25) μMolars curcumin intended for 24h. After cultured and treatment cells proliferation and viability were detected by MTT assay. The expression levels of these proteins (p53, TIGAR, Beclin-1 and LC3) were analyzed by immunoblotting assay and the expression of LC3 protein autophagy marker were analyzed by immunocytochemistry assay. The MTT assay results demonstrated that curcumin lead to distinct suppression of osteosarcoma cell proliferation and cell viability. There were observed a linear dose/depends effect for curcumin range 15-25 μM dose cause to modulate of p53, TIGAR, Beclin1 and LC3. Analysed protein reveals a relation reduction in the levels of p53 and TIGAR proteins above treatment with 10–25 μM curcumin. The result of immunocytochemistry showed that by increasing curcumin dose significantly increased the expression of the total amount LC3 protein in cytoplasm osteosarcoma U2OS cells. Our studied has improved the perceptive of the fundamental technique anticancer properties of curcumin.; ÖZET:&#13;
Osteosarkom, kemiklerde en sık rastlanan primer tümörlerdir ve genellikle tibia, femur, humerus gibi uzun boru şekilli kemiklerin metafizi çevresinde ortaya çıkar. Curcumin, Curcuma longa L'nin kurutulmuş toprak rizomlarındaki en önemli etkiye sahip bileşendir. Yara iyileşmesini destekler, karaciğer hasarına, katarakt oluşumuna ve pulmoner toksisiteye karşı koruma sağlar. Curcumin (zerdeçal) geleneksel Çin tıbbında binlerce yıldır kullanılmaktadır. Anti-bakteriyel, anti-inflamatuvar, anti-oksidan ve anti-kanser özellikleri de dahil olmak üzere birçok farmakolojik etkiue sahip olduğu bildirilmiştir. Curcumin’in, hücre invazyonunu engellemek, migrasyonu ve inflamatuar yanıtları bastırmak, apoptoz veya otofajiyi tetiklemek birçok etkisinin olduğu kanıtlanmıştır. Kansere karşı curcumin etkilerinin kapsamlı bir şekilde araştırılmış olmasına rağmen curcumin'in p53 / TIGAR ve otofajik yolak üzerindeki etkisi U2OS hücre hattında henüz ele alınmamıştır. Bu nedenle insan osteosarkom U2OS hücre hattında curcuminin p53 / TIGAR ve otofaji yolu üzerindeki etkisininin araştırılması amaçlanmıştır. Osteosarkom U2OS hücreleri 24 saat süreyle farklı konsantrasyonlarda (10, 15, 20 ve 25 μM) curcumin ile tedavi edildi. Hücre çoğalması hücre canlılık testi olan MTT ile saptanmıştır. Bu proteinlerin (p53, TIGAR, Beclin-1 ve LC3) ekspresyon seviyeleri, western blotting ile analiz edilmiştir. MTT analizinin sonuçları, curcumin'in osteosarkom hücre proliferasyonunun ve hücre canlılığının inhibisyonuna neden olduğunu göstermiştir. Curcumin aralığı 15-25 μM doz ve p53, TIGAR, Beclin1 ve LC3 modülasyonu için doza / bağımlı doğrusal bir etkisinin olduğu belirlenmiştir. Osteosarkom U2OS hücre hattı 15-25 uM curcumin ile tedavi edildiğinde p53 ve TIGAR protein seviyelerinde bir düşüş olduğu ortaya konmuştur. Beclin-1'in curcumin tarafından inhibe edilmesi otofajiye spesifik belirteç olan mikrotübüle bağlı protein LC3’nin artmasına yol açmaktadır. Immünositokimya sonucunda curcumin dozunun artmasıyla total LC3 proteininin ekspresyonunun belirgin şekilde arttığı gösterilmiştir. Bu bulgular, curcumin'in antikanser özelliklerinin altında yatan mekanizmanın anlaşılmasını geliştirmiştir.

Non-small cell lung cancer has a high mortality rate with a lethal malignancy. siRNA is the most-commonly used RNA interference tool for inducing short-term silencing of protein-coding genes in the post-transcriptional process as negative regulators of gene expression. The TIGAR protein controls Fru-2, 6-P2 levels during glucose metabolism and helps maintain NADPH levels to recycle GSH, a key intracellular antioxidant. One of TIGAR’s functions is to lower Fru-2, 6-P2 levels in cells, consequently decreasing glycolysis and suppressing ROS, which correlate with higher resistance to cell death. However, little is known about the effects of siRNA on cell viability, oxidative stress, and programmed cell deaths, or about how siRNA interacts with TIGAR in lung cancer cells. The present study was designed to investigate the molecular mechanisms underlying TIGAR knockdown in the A549 cell line.&#13;
To detect the influence of siRNA-TIGAR on A549 lung cancer cells, we performed cell proliferation, colony formation, ROS, and NADPH assays to measure cell viability, ability of cells to form colonies, and oxidative stress levels of ROS and NADPH, respectively. In addition, western blotting and real-time PCR assays were used to measure protein and mRNA expression levels, respectively.&#13;
After TIGAR-knockdown in A549 cell lines, various assay parameters were analyzed and showed that down regulation of TIGAR inhibited viability and decreased colony formation. We also demonstrated that TIGAR knockdown induced apoptosis and autophagy. Furthermore, increased ROS levels and decreased NADPH levels were observed, followed by an induction of oxidative stress marker expression. In summary, siRNA TIGAR inhibited cell viability, enhanced programmed cell deaths (I, II), and elevated oxidative stress markers by increasing ROS levels.&#13;
The data showed that TIGAR knockdown induced inhibition of cell viability; increased ROS levels; and decreased NADPH levels, induction of autophagy and apoptosis, and oxidative stress markers. Overall, this study supports our understanding the possibility of using TIGAR knockdown in lung cancer cells to enhance programmed cell death I, II for further use as a therapeutic target in cancer treatment.; ÖZET:&#13;
Küçük hücreli olmayan akciğer kanseri ölümcül bir malignite ile yüksek mortalite oranına sahiptir. siRNA, gen ifadesinin negatif düzenleyicileri olarak transkripsiyon sonrası süreçte protein kodlayan genlerin kısa süreli susturulmasını indüklemek için en yaygın kullanılan RNA interferens aracıdır. TIGAR proteini,glikoz metabolizması sırasında Fru-2, 6-P2 seviyelerini kontrol eder ve NADPH düzeyini devam ettirerek hücre içi önemli birantioksidan olan GSH'nin geri dönüştürmesine yardımcı olur. TIGAR'ın işlevlerinden biri, hücrelerdeki Fru-2, 6-P2 seviyelerini düşürmek ve sonuç olarak hücre ölümüne karşı daha yüksek dirençle korele olan ROS'u baskılamak ve glikolizi azaltmaktır. Bununla birlikte, siRNA'nın hücre canlılığı, oksidatif stres ve programlanmış hücre ölümleri üzerindeki etkileri veya siRNA'nın akciğer kanseri hücrelerinde TIGAR ile nasıl etkileştiği hakkında pek az şey bilinmektedir. Bu çalışma, A549 hücre hattında TIGAR'ın susturulmasının altında yatan moleküler mekanizmaları araştırmak için tasarlanmıştır.siRNA-TIGAR'ın A549 akciğer kanseri hücreleri üzerindeki etkisini saptamak için hücre canlılığını, koloni oluşumunu, ROS ve NADPH'nin oksidatif stres seviyelerini ve hücre canlılığını ölçmek için sırasıyla hücre proliferasyonu, koloni oluşumu, ROS ve NADPH analizleri gerçekleştirdik. Ek olarak, protein ve mRNA ekspresyon seviyelerini ölçmek için sırasıyla western blot ve real-time PCR analizleri kullanıldı.A549 hücre hattında TIGAR inaktif edildikten sonra çeşitli parametreler analiz edildi ve TIGAR'ın down regülasyonunun canlılığı inhibe ettiği ve koloni oluşumunu azalttığı gösterildi. Ayrıca TIGAR inaktivasyonunun apoptozis ve otofajiyi tetiklediği görüldü. Artmış ROS seviyeleri ve azalmış NADPH seviyelerine bağlı olarak oksidatif stres belirteçlerinin indüksiyonu gözlemlendi. Özetle, siRNA TIGAR hücre canlılığını inhibe edip programlanmış hücre ölümlerini (I, II) arttırmıştır ve ROS seviyesini artırarak yüksek oksidatif stres markırlarını yüksek düzeylere çıkarmıştır. Bulgular, TIGAR’ı susturmanın hücre canlılığı inhibisyonunu indüklediğini, ROS seviyelerini artırdığını, NADPH düzeylerini azalttığını, otofaji, apoptosis ve oksidatif stres belirteçlerini indüklediğini gösterdi. Genel olarak, bu çalışma kanser tedavisinde terapötik bir hedef olarak kullanılmak üzere akciğer kanseri hücrelerinde programlanmış hücre ölümü I, II'yi arttırmak için TIGAR susturmanın muhtemel rolünün anlaşılmasını desteklemektedir.

Bu çalışmada 2014-2016 yılları arasında Bingöl ili sınırları içerisinden toplanan Harpalinae (Coloeptera: Carabidae) altfamilyasına ait türler, faunustik ve sistematik açıdan araştırılmıştır. Araştırma alanından çukur tuzaklar ve el ile toplanan 550 örneğin incelenmesi sonucunda Harpalinae altfamilyasına ait 4 cins ve 13 tür Acinopus (Acinopus) laevigatus Ménétriés, 1832; Acinopus (Acinopus) picipes (Olivier, 1795); Dixus (Dixus) eremita (Dejean, 1825); Dixus (Dixus) obscurus (Dejean, 1825); Harpalus (Harpalus) affinis (Schrank, 1781); Harpalus (Harpalus) attenuatus Stephens, 1828; Harpalus (Harpalus) caspius (Steven, 1806); Harpalus (Harpalophonus) hospes (Sturm, 1818); Harpalus (Harpalus) latus (Linnaeus, 1758); Harpalus (Harpalus) serripes (Quensel, 1806); Harpalus (Harpalus) smaragdinus (Duftschmid, 1812); Ophonus (Hesperophonus) cribricollis (Dejean, 1829); Ophonus (Metophonus) puncticollis (Paykull,1798) tespit edilmiştir.&#13;

In this study, the species belonging to Harpalinae (Coloeptera: Carabidae) subfamily collected from Bingöl province between 2014-2016 were investigated in terms of faunoustics and systematic aspects. 13 species Acinopus (Acinopus) laevigatus Ménétriés, 1832; Acinopus (Acinopus) picipes (Olivier, 1795); Dixus (Dixus) eremita (Dejean, 1825); belonging to Harpalinae subfamily and collected by hand traps and pit traps (550 samples from the study area).
