Farklı Fesleğen (Ocimum Basilicum L.) Genotiplerinin Çimlenme Özelliklerini Etkileyen Polietilen Glikol Dozlarının Belirlenmesi

Abstract
ÖZET: Fesleğen bitkisinin kuraklık stresinde çimlenmesi, çıkışı ve fide gelişim evrelerinde dayanaklılığının artırılması, önemli bir unsur olarak bilinmektedir. Bu bitkide gelişim aşamaları arasında, çimlenme ve fide gelişimlerinin olduğu dönem, strese en hassas oldukları dönem olduğundan dolayı bu çalışmada Türkiye’de bulunan farklı fesleğen hatlarının kuraklığa dayanıklılığı incelenmiş olup, tohumların kuraklık streslerinde çimlenme, çıkış ve fide gelişim dönemlerindeki performansları karşılaştırılmıştır. Deneme Bingöl Üniversitesi Ziraat Fakültesi Tarla Bitkileri Bölümü Laboratuvarlarında, faktöriyel çalışması olarak, tesadüf parselleri deneme desenine göre üç tekrarda yürütülmüştür. Araştırmada, beş farklı popülasyon (Elâzığ, Kahramanmaraş, Hatay, Denizli ve İstanbul) ve beş farklı PEG 6000 konsantrasyon (kuraklık stresi oluşturmak için %0,0- %2,5- %5,0- %7,5 ve %100 oranında PEG) kullanılmış olup, tohum çimlenmesi ve fide büyümesine olan etkileri araştırılmıştır. Elde edilen veriler ışığında, Ocimum basilicum L. genotiplerinin abiyotik stres koşullarına farklı tepkiler verdiği belirlenmiştir. Kuraklık stresinde Ocimum basilicum L. tohumlarının Elâzığ ve Hatay popülasyonları daha iyi sonuçlar verirken, en az ve en çok çimlenme yüzdesi sırasıyla %0 PEG ve %100 PEG dozlarında görülmüştür. Ayrıca kuraklık stresinde fide uzunluğu bakımından Denizli ve Elâzığ popülasyonları sırasıyla en az ve en çok fide uzunluğuna sahip popülasyonlar olarak belirlenmiştir. Sonuç olarak, Elâzığ, Hatay ve Denizli popülasyonları çimlenme ve fide gelişim dönemlerinde, düşük PEG streslerine toleranslı olduğu ve bu amaçla ıslah çalışmalarında dikkate alınması gereken çeşitler olabileceği düşünülmektedir.
 
ABSTRACT: It is known as an important factor to increase the resilience of the basil plant in drought stress, germination, emergence and seedling development stages. Since the germination and seedling development periods are the most sensitive to stress among the development periods of this plant, drought resistance of different basil lines in Turkiye was investigated in this study and the performances of the seeds in the germination, emergence and seedling development periods were compared under drought stress. The experiment was carried out in the laboratories of the Department of Field Crops, Faculty of Agriculture, Bingöl University, in three repetitions as a factorial study, according to the randomized plot design. In the study, five different populations (Elazig, Kahramanmaraş, Hatay, Denizli and Istanbul) and five different PEG 6000 concentrations (0,0%- 2,5%- 5,0- 7,5% and 10,0% PEG to create drought stress) were used to improve seed germination and the effects on seedling growth were investigated. According to the obtained data, it was determined that basil genotypes gave different responses to abiotic stress conditions. Ocimum basilicum L. seeds gave better results in Elâzığ and Hatay populations under drought stress, while the minimum and maximum germination percentages were observed at 0% PEG and 100% PEG doses, respectively. In addition, in terms of seedling length in drought stress, Denizli and Elazig populations were determined as the populations with the lowest and highest seedling length, respectively. As a result, it is thought that Elazig, Hatay and Denizli populations were tolerant to low PEG stresses during germination and seedling development periods and may be varieties to be considered in breeding studies for this purpose.
