ÖZET:&#13;
Nörodejeneratif hastalıkların gelişiminde oksidatif stresin önemli bir rol oynaması nedeniyle, biyouyumlu ve çevre dostu nanomalzemelerin geliştirilmesi güncel araştırma alanları arasında yer almaktadır. Bu çalışmada, kahve posası ekstraktı kullanılarak yeşil sentez yöntemi ile gümüş nanopartikülleri (KP@Ag NPs) sentezlenmiş ve elde edilen nanopartiküller kitosan/tripolifosfat (TPP) esaslı nanojel sistemine yüklenerek CS@KP@Ag nanojel formülasyonu geliştirilmiştir. Sentezlenen nanopartiküller ve nanojel sisteminin fizikokimyasal özellikleri UV-Vis spektrofotometrisi, FT-IR, LC-MS/MS, dinamik ışık saçılımı (DLS), zeta potansiyeli, taramalı elektron mikroskobu (SEM) ve enerji dağılımlı X-ışını spektroskopisi (EDX) analizleri ile karakterize edilmiştir. Biyolojik etkinlik çalışmaları, H₂O₂ ile oksidatif stres oluşturulan SH-SY5Y insan nöroblastoma hücre hattında gerçekleştirilmiş; hücre canlılığı WST yöntemi ile değerlendirilirken toplam antioksidan seviye (TAS) ve toplam oksidan seviye (TOS) analizleri ile antioksidan etkinlik araştırılmıştır. Karakterizasyon sonuçları, gümüş nanopartiküllerinin başarıyla sentezlendiğini ve nanojel matrisi içerisine etkin şekilde yüklendiğini göstermiştir. Ayrıca CS@KP@Ag nanojel uygulamasının oksidatif stres koşullarında hücre canlılığını artırdığı, toplam antioksidan seviyesini yükselttiği ve toplam oksidan seviyesini azalttığı belirlenmiştir. Sonuç olarak, kahve posasının katma değerli bir biyomateryale dönüştürülmesiyle geliştirilen CS@KP@Ag nanojel sisteminin başarılı fizikokimyasal özelliklere sahip olduğu, oksidatif strese karşı nöroprotektif potansiyel taşıdığı ve biyomedikal ile farmasötik uygulamalarda kullanılabilecek çevre dostu ve umut vadeden bir nano-taşıyıcı sistem olduğu değerlendirilmiştir.; ABSTRACT:&#13;
Oxidative stress plays a critical role in the development of neurodegenerative diseases, making the development of biocompatible and environmentally friendly nanomaterials an important area of research. In this study, silver nanoparticles (KP@Ag NP) were synthesized using a green synthesis approach with spent coffee ground extract, and the synthesized nanoparticles were incorporated into a chitosan/tripolyphosphate (TPP)-based nanogel system to develop a CS@KP@Ag nanogel formulation. The physicochemical properties of the synthesized nanoparticles and nanogel system were characterized using UV-Vis spectroscopy, Fourier-transform infrared spectroscopy (FT-IR), liquid chromatography–tandem mass spectrometry (LC-MS/MS), dynamic light scattering (DLS), zeta potential analysis, scanning electron microscopy (SEM), and energy-dispersive X-ray spectroscopy (EDX). The biological activity of the developed formulation was evaluated in H₂O₂-induced oxidative stress model using the SH-SY5Y human neuroblastoma cell line. Cell viability was determined by the WST assay, while antioxidant activity was assessed by measuring total antioxidant status (TAS) and total oxidant status (TOS). Characterization results confirmed the successful synthesis of silver nanoparticles and their efficient incorporation into the nanogel matrix. Furthermore, treatment with the CS@KP@Ag nanogel significantly increased cell viability under oxidative stress conditions, enhanced total antioxidant status, and reduced total oxidant status. These findings suggest that the developed CS@KP@Ag nanogel effectively alleviates oxidative stress-induced cellular damage and exhibits promising neuroprotective potential. In conclusion, the CS@KP@Ag nanogel, developed by valorizing spent coffee grounds into a value-added biomaterial, possesses favorable physicochemical properties and demonstrates considerable biological activity, indicating its potential as an environmentally friendly and promising nanocarrier system for future biomedical and pharmaceutical applications.

SARUHAN, Seher
ÖZET:&#13;
Akciğer kanseri, dünya çapında birçok kanser arasında en sık görülen kanser türüdür. PFKFB3, glikoliz sürecinde önemli role sahip bir enzimdir. Sahip olduğu yüksek kinaz/fosfataz oranı ile (740:1) kanser hücrelerinde önemli bir hedef olarak kabul edilmektedir. Bu bağlamda yapılan çalışmalar ışığında geliştirilen PFKFB3 hedefli PFK-158 küçük molekül inhibitörü ile solid tümör hastalarında faz I klinik çalışmalar gerçekleştirilmiştir. Söz konusu çalışmaların neticesinde, PFKFB3’ü hedefleyen, daha etkin bir küçük molekül inhibitörlerinin geliştirilmesi amaçlanmıştır. Yakın zaman önce ortaya çıkan KAN0438757 inhibitörü, normal hücrelere zarar vermeden transforme hücrelerde çoğalmayı inhibe ettiği bildirilmiştir. Resveratrol bileşiği üzüm, kırmızı şarap, yer fıstığı, asma yapraklarında bulunur ve patojenler bitkilere saldırdığında ve yaralandığında bitkiler tarafından üretilir. Ayrıca resveratrol antikanser etkilere sahiptir ve hücre büyümesi, inflamasyon, apoptoz, metastaz ve anjiyogenezi kontrol ederek çeşitli sinyal iletim yollarını etkileyerek kanserin tüm evrelerini etkilemektedir. Bu çalışmada, resveratrol ve KAN0438757 kombinasyonunun, hücre proliferasyonu, hücre ölümü, mitokondriyal membran potansiyeli, hücre göçü ve programlı hücre ölümü biyobelirteçleri üzerine etkilerinin belirlenmesi amaçlanmaktadır. Elde edilen veriler ile resveratrol ve KAN0438757 kombinasyon tedavisinin hücre canlılığını önemli ölçüde inhibe ettiği ve hücre proliferasyonunu azalttığı gösterilmiştir. Aynı zamanda kombinasyon tedavisi ile hücre göçünün engellendiği ve hücrelerin programlı hücre ölümü ile apoptoza gittiği tespit edilmiştir. Elde edilen bu veriler, resveratrolün, KAN0438757’nin etkinliğini arttırdığını ortaya koymaktadır.; ABSTRACT:&#13;
Lung cancer is the most common type of cancer among many cancers worldwide. PFKFB3 is an enzyme that has an important role in the glycolysis process. With its high kinase/phosphatase ratio (740:1), it is considered an important target in cancer cells. Phase I clinical studies were carried out in solid tumor patients with the PFKFB3-targeted PFK-158 small molecule inhibitor, which was developed in the light of the studies conducted in this context. As a result of these studies, it was aimed to develop more effective small molecule inhibitors targeting PFKFB3. The recently emerged inhibitor KAN0438757 has been reported to inhibit proliferation in transformed cells without harming normal cells. Resveratrol compound is found in grape, red wine, peanut, vine leaves and is produced by plants when pathogens attack and injure plants. In addition, resveratrol has anticancer effects and affects all stages of cancer by affecting various signal transduction pathways by controlling cell growth, inflammation, apoptosis, metastasis and angiogenesis. In this study, it is aimed to determine the effects of resveratrol and KAN0438757 combination on cell proliferation, cell death, mitochondrial membrane potential, cell migration and programmed cell death biomarkers. With the obtained data, it has been shown that resveratrol and KAN0438757 combination therapy significantly inhibited cell viability and reduced cell proliferation. At the same time, it was determined that cell migration was inhibited by combination therapy and cells went to apoptosis with programmed cell death. These data reveal that resveratrol increases the effectiveness of KAN0438757.

<title>Investigation of the Effect of Reduced MOB2 Expression on the Olaparib Response of PARP1/2 Inhibitor-Resistant Cancer Cells</title>

<description>Investigation of the Effect of Reduced MOB2 Expression on the Olaparib Response of PARP1/2 Inhibitor-Resistant Cancer Cells
AÇIKGÖZ, Sıdıka
ABSTRACT:&#13;
PARP enzymes play significant roles in various cellular processes, including DNA single-strand break repair. Inhibition of PARP activity results in the accumulation of DNA single-strand breaks, which are eventually converted to DNA double-strand breaks, especially during DNA replication. Cells with deficient homologous recombination DNA double-strand break repair pathway are hypersensitive to PARP inhibitors due to deleterious accumulation of DNA breaks. Although several PARP inhibitors have been effectively used in the clinic, PARP inhibitor resistance that ultimately develops in most patients limits clinical success. Therefore, identification of pharmacologically important cellular targets and predictive biomarkers may ameliorate the current treatment strategies with PARP inhibitors. hMOB2 protein is a potential DNA damage response regulator that supports homologous recombination repair mechanism. hMOB2-defective cancer cells, thus, become sensitive to PARP inhibition. In this research, it was investigated whether hMOB2 deficiency may also render PARP inhibitor-resistant cancer cells selectively sensitive towards PARP inhibition. Mammalian cell culture and various molecular biology techniques were used to dissect our research questions. U2OS osteosarcoma cancer cells and olaparib, a potent PARP inhibitor widely used in the clinic, were employed in our experiments. U2OS cells were subjected to continuously increased doses of olaparib to establish PARP inhibitor-resistant clones (U2OS/PIR cells). Interestingly, p21 protein levels were dramatically elevated in U2OS/PIR cells compared to parental counterparts, although no difference was observed in p53 and hMOB2 expressions. Notably, hMOB2 silencing significantly reduced the survival of U2OS/PIR cells in the absence of olaparib. Dual inhibition of hMOB2 and PARP activities further sensitized resistant cancer cells towards olaparib treatment. Taken together, our experiments revealed that, against PARP inhibition, hMOB2 protein is likely to be an important factor that supports not only parental cells but also PARP inhibitor-resistant cells to survive.; ÖZET:&#13;

<title>C60 Fulleren Nanoparçacıkların Glioblastom U373 Hücreleri Gelişimi ve İstilası Üzerindeki Etkisi</title>

<description>C60 Fulleren Nanoparçacıkların Glioblastom U373 Hücreleri Gelişimi ve İstilası Üzerindeki Etkisi
BAYSAL, Sevda
ÖZET:&#13;
Glioblastoma Multiforme (GBM), birincil Merkezi Sinir Sistemi tümörü olup, tüm beyin tümörleri arasındaki en sık görülen, en agresif ve en ölümcül olanıdır. Tedavi stratejileri arasında cerrahi tedavi, radyoterapi ve kemoterapi bulunmaktadır. Ancak GBM hücrelerinin istilaya eğilimli olmaları nedeniyle bu tedavi stratejileri yetersiz kalmaktadır. Bu nedenle yeni tedavi stratejileri araştırılmakta ve özellikle de hedefe yönelik ilaç tedavi yöntemleri üzerinde durulmaktadır. Tam olarak bu noktada nanoteknoloji kullanılmaktadır. Nanoteknoloji ile üretilen nanoparçacıklar ve özellikle C60 fulleren güçlü antikanser aktivite, güçlü antioksidan etki, antimikrobiyal etki, antiviral etki ve radikal süpürücü özellikleri ile GBM tedavisinde hem hedefe yönelik ilaç hem de yeni bir teknoloji olarak kullanılmaktadır. Bu çalışmanın amacı, nanopartiküllerin kanser hücreleri üzerindeki etkisinin mekanizmalarını netleştirmek için glioblastoma U373 hücrelerinde C60 fulleren’in istila aktivitesi ve hücre iskeleti proteinlerinin ekspresyonu üzerindeki etkisinin incelenmesidir. Mevcut çalışmanın sonucunda, C60 fulleren’in glioblastoma U373 hücresinin özellikle hücre iskeletini, fibronektin üretimini ve hareketliliğini etkilemek için güçlü olduğunu göstermiştir. Ayrıca mevcut çalışmanın sonuçlarında gözlenen veriler bir bütün halinde değerlendirildiğinde C60 fulleren, hücre iskeleti ana yapıları olan ara filamentleri ve mikrotübülleri zıt yönde etkilemiştir. Bununla birlikte, C60 fulleren’in glioblastoma hücre göçü üzerindeki inhibitör etkisine, GFAP yukarı regülasyonu ve Tübülin aşağı regülasyonu eşlik ettiği tespit edilmiştir. C60 fulleren nanoparçacıkları, hücre iskeletinin yeniden düzenlenmesi üzerindeki etkinin yanı sıra glioblastoma hücrelerinin reaktivitesini de düzenler. Bu nedenle C60 fulleren, GBM istilasında rol oynadığı gibi glioblastoma hareketliliğinin baskılanmasında da rol oynayabilir.; ABSTRACT:&#13;
Glioblastoma Multiforme (GBM) is the primary Central Nervous System tumor and is the most common, most aggressive and deadly of all brain tumors. Treatment strategies include surgery, radiotherapy, and chemotherapy. However, these treatment strategies are insufficient because GBM cells are prone to invasion. For this reason, new treatment strategies are being researched and especially targeted drug treatment methods are emphasized. This is exactly where nanotechnology is used. Nanoparticles produced with nanotechnology and especially C60 fullerene are used as both a targeted drug and a new technology in GBM treatment with their strong anticancer activity, strong antioxidant effect, antimicrobial effect, antiviral effect and radical scavenging properties. The aim of this study was to examine the effect of C60 fullerene on the invasion activity and expression of cytoskeletal proteins in glioblastoma U373 cells to clarify the mechanisms of the effect of nanoparticles on cancer cells. The result of the present study showed that C60 fullerene is particularly potent to affect the cytoskeleton, fibronectin production and motility of glioblastoma U373 cell. In addition, when the data observed in the results of the current study were evaluated as a whole, C60 fullerene affected the intermediate filaments and microtubules, which are the main cytoskeletal structures, in the opposite direction. However, the inhibitory effect of C60 fullerene on glioblastoma cell migration was found to be accompanied by GFAP upregulation and Tubulin downregulation. C60 fullerene nanoparticles regulate the reactivity of glioblastoma cells as well as the effect on cytoskeletal reorganization. Therefore, C60 fullerene may play a role in the suppression of glioblastoma motility as well as in GBM invasion.
