Aurora B Kinaz İnhibitörü BI 831266 Molekülünün Sisplatin Tedavisinin Sitotoksik Potansiyeli Üzerine Olan Etkisinin Belirlenmesi

Özet
ÖZET: Kanser, proteinlerin işlevsizliği nedeniyle hücrelerin kontrolsüz bir şekilde çoğalması sonucunda vücutta meydana gelen tedavi edilmesi zor bir hastalıktır. Kanser tedavisinde kullanılan konvansiyonel yöntemler başarılı sonuçlar sağlasa da bu tedaviler hastalar üzerinde ciddi yan etkilere ve bir süre sonra hücrelerin bu tedavilere direnç geliştirmesine neden olmaktadır. Bu önemli nedenlerden dolayı kişiselleştirilmiş tedaviler oldukça önem arz etmektedir. Kemoterapide hali hazırda kullanılan ilaçlarla küçük molekül inhibitörlerin kombinasyon tedavileri sinerjistik etki yaratarak bu olumsuz etkileri ortadan kaldırmak için önemli rol oynamakta ve küçük molekül inhibitörleri önemli kılmaktadır. Bu çalışmada, hücre döngüsünün önemli bir regülatörü olan Aurora B kinaz inhibisyonunun konvansiyonel anti-kanser tedavisinde başarılı bir şekilde kullanılan sisplatinin sitotoksisitesi üzerine olan etkisi araştırıldı. Aurora B inhibisyonu için daha önce üzerine herhangi bir kombine sitotoksik analiz yapılmamış olan BI 831266 molekülü kullanıldı. Sisplatin ve BI 831266 molekülünün mono ve kombine tedavi sitotoksik etkileri insan meme kanseri hücrelerinde (MCF-7) incelendi. MCF-7 hücreleri artan dozlarda sisplatin ve BI 831266 molekülü ile tedavi edildi ve sitotoksik etki koloni sağ-kalım analiziyle belirlendi. IC50 dozları belirlenen sisplatin ve BI 831266 molekülünün sinerjistik etkisi kombine tedavi ile belirlendi. Sisplatin ve BI 831266 molekülünün kombinasyon tedavisi, koloni sağ-kalım analizi ile kanser hücrelerinin koloni oluşma kapasitelerini inhibe ettiği gösterildi. Sisplatin ve BI 831266 molekülünün mono ve kombinasyon tedavileri MCF-7 hücrelerinde DNA hasarı ve apoptotik sinyal yolaklarındaki proteinlerin ekspresyon düzeylerine western blot yöntemiyle bakıldı. Bu çalışmada elde edilen veriler, sisplatin ve BI 831266 molekülünün kombinasyon tedavileri hücre canlılığını azalttığı ve koloni sağ-kalımlarını baskıladığı, apoptozisle ilişkili proteinlerin ekspresyon seviyelerini regüle ederek DNA hasarına ve apoptoza sebep olduğunu göstermektedir.
 
ABSTRACT: Cancer is a difficult to treat disease that occurs in the body as a result of the uncontrolled proliferation of cells due to the dysfunction of proteins. Although conventional methods used in cancer treatment provide successful results, these treatments cause serious side effects on patients and after a while, the cells develop resistance to these treatments. For these important reasons, personalized treatments are very important. Combination treatments of drugs currently used in chemotherapy and small molecule inhibitors play an important role in eliminating these negative effects by creating a synergistic effect, making small molecule inhibitors important. In this study, the effect of inhibition of Aurora B kinase, an important regulator of the cell cycle, on the cytotoxicity of cisplatin, which has been used successfully in conventional anti-cancer therapy, was investigated. For Aurora B inhibition, BI 831266 molecule, on which no combined cytotoxic analysis was performed before, was used. The mono- and combined-therapy cytotoxic effects of cisplatin and BI 831266 were investigated in human breast cancer cells (MCF-7). MCF-7 cells were treated with increasing doses of cisplatin and the BI 831266 molecule, and the cytotoxic effect was determined by colony survival analysis. The synergistic effect of cisplatin and BI 831266 molecule, whose IC50 doses were determined, was determined by combined treatment. Combination treatment of cisplatin and BI 831266 molecule was shown to inhibit colony formation capacity of cancer cells by colony survival assay. Mono and combination treatments of cisplatin and BI 831266 molecule DNA damage and expression levels of proteins in apoptotic signaling pathways in MCF-7 cells were evaluated by western blot method. The data obtained in this study show that combination treatments of cisplatin and BI 831266 molecule reduce cell viability and suppress colony survival, and cause DNA damage and apoptosis by regulating the expression levels of apoptosis-related proteins.
