Abstract

ABSTRACT: Propolis stands out as an important natural ingredient in the development of functional foods due to its high phenolic compound content and strong antioxidant and antimicrobial properties. However, its intense aroma, low water solubility and stability issues limit its use in food systems. In this thesis, two innovative approaches were evaluated to increase the applicability of propolis to different food matrices. In the first approach, propolis was encapsulated in a chitosan-based colloidal delivery system and applied to walnut sucuk, a traditional product; in the second approach, propolis and bee pollen were included in a kombucha fermentation system and their effects on functional properties were investigated. In addition, pollen was encapsulated in a chitosan Pickering emulsion and the potential of chitosan in the transport of pollen extract was investigated. Different chitosan concentrations and DATEM/lecithin combinations were used in the preparation of colloidal systems. Structural characterizations confirmed that propolis was successfully loaded in the chitosan colloid system. It was determined that the encapsulation efficiency of phenolic compounds varied depending on their chemical structure, with chrysin, vanillin and benzoic acid showing particularly high encapsulation efficiency. The addition of propolis loaded colloid to the walnut sausage formulation increased the TPC and antioxidant capacity while significantly reducing lipid oxidation and revealed that the chitosan colloidal system enabled the efficient transport of propolis into the food matrix. In the kombucha study, the effects of propolis, bee pollen and their combinations on the fermentation process and the physicochemical, microbiological, sensory and functional properties of the product were investigated. It was determined that bee products did not negatively affect the development of the SCOBY microbiota and that fermentation was successfully carried out. While the addition of propolis significantly increased the total phenolic content, the combined use of propolis and bee pollen resulted in the highest antioxidant capacity. UHPLC-Orbitrap®-HRMS analyses showed that bee products significantly altered the phenolic composition and amino acid profile of kombucha. Samples with added propolis were enriched in hydroxycinnamic acid derivatives and aglycone flavonoids, while samples with added pollen were enriched in flavonoid glycosides and amino acids. Furthermore, the addition of bee pollen enhanced sensory acceptance and supported microbial growth. These findings contribute to the development of new carrier systems for increasing the stability and preserving the functional properties of natural bioactive components, while providing a scientific basis for evaluating bee products as innovative functional components in different food matrices.
