ABSTRACT: Glutathione S-transferase enzyme (GST; EC 2.5.1.18) is an important antioxidant enzyme in metabolism and takes part in reactions that provide conjugation of glutathione with many metabolites that may cause toxicity. Within the scope of this thesis, the GST enzyme, which has 9,048 EU/mg protein specific activity, was purified 20.99-fold with 40% yield by means of preparation of homogenate from chicken heart, ammonium sulfate precipitation and glutathione-agarose affinity chromatography. Then, sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) was performed to control the enzyme purity and to determine the subunit molecular mass, and a single band was obtained. The subunit molecular mass of the GST enzyme purified by SDS-PAGE was found to be approximately 20.94 kDa. In the characterization studies for the additionally purified GST enzyme, optimum pH (in Tris-HCl buffer) pH= 8.5, optimum ionic strength (in Tris-HCl buffer) 0.2 M, stable pH, (in Tris-HCl buffer) pH = 8, 5 and the optimum temperature is 60 oC; also, using Lineweaver-Burk graphs, the KM and Vmax values of the enzyme were determined as KM, 0.087 mM, Vmax 0.625 EÜ/mL for the GSH substrate; For CDNB substrate, KM was calculated as 0.798 mM, Vmax was calculated as 1.141 EU/mL.
